Recombinant Rabbit Monoclonal Antibody Generation

Rabbit antibody phage display library has many advantages like it can produce monoclonal antibodies with high specificity, high activity and high affinity in a short time. Therefore, rabbit antibody phage display library is a good source of abbit-derived polyclonal antibodies and monoclonal antibodies. Compared with hybridoma technology, hybridomas prepared by phage library technology can be stored on phagemid vectors in the form of antibody genes, combining the advantages of different protein expression systems, such as the use of cell-free expression systems, enabling animal-free production and obtaining high specificity and high affinity recombinant monoclonal antibody. The ability of rabbit monoclonal antibodies to recognize the common antigenic determinants of rodents and human antigens is much better than that of murine monoclonal antibodies, which is also due to the distant evolution of rat and mouse commonly used in rabbits and rodents. Rodent monoclonal antibodies have no effect on the common antigenic determinants of rodents and human antigens, while rabbit antibodies often recognize the common antigenic determinants of rodents and human antigens, overcoming this obstacle in antibody production.

In the process of developing recombinant monoclonal antibody by phage technology, KMD Bioscience utilizes rich experience and technology accumulation to construct chimeric antibody by cloning rabbit antibody variable region (Vλ, Vk, and VH) into human antibody backbone monovalent protein to avoid the toxicity of rabbit Fab antibody fragments to E. coli. Subsequently, the highly specific and high affinity rabbit-derived antibody variable regions (Vλ, Vk, and VH) were cloned into a plasmid vector carrying a rabbit antibody scaffold protein. Combined with our recombinant protein expression platform the yield can be up to 5g/L with our transient expression.
