During the past few decades, different techniques have been developed to produce monoclonal antibodies. Compared with mouse monoclonal antibodies, rabbit monoclonal antibody is widely used in scientific research because of their high affinity and abundant epitopes. Based on the single B cell cloning technique, positive B cells can be obtained from peripheral blood directly after animal immunization and antibody gene sequences can be cloned. Step1: Rabbit Immunization

* A Rabbit was immunized 4-7 times

* Serum titers were measured by ELISA and collected peripheral blood

Step2: B cell enrichment and screening

* Collection of lymphocytes or PBMCs by density gradient centrifugation from a few milliliters of animal blood

* Fluorescent staining of plasma cells with endoplasmic reticulum (ER)-tracker

* Sorting of stained cells by fluorescence-activated cell sorting (FACS)

* Adsorption of cells binding to the antigen using antigen-coated magnetic beads

* Separation of one cell per 10 µL into a 384-well plate

* Selection and confirmation of cell-bead complexes under an inverted phase-contrast microscope

Step3: Single B Cell Sequencing

* Reverse transcription using gene-specific primers and SuperScript IV reverse transcriptase

* First PCR, using primers annealing to the signal peptide (SP) sequence and constant region of antibody genes

* Second PCR, using primers with tails required for the subsequent Gibson assembly step

* Gibson assembly to combine T7 promoter and terminator with antibody genes

* PCR to amplify DNA fragments for cell-free protein synthesis. HA and His tags are present downstream of leucine zipper A (LZA) and B (LZB), respectively
