PDCD1/CD279 Target

CD279 (Gene ID:5133) was discovered in the late 1990s as a member of the CD28/CTLA-4 family of immune checkpoint receptors. In contrast to CD28, which provides co-stimulatory signals for T-cell activation, CD279 transmits inhibitory signals upon binding to its ligands, PD-L1 (B7-H1) and PD-L2 (B7-DC). It is expressed in various cell types, including cancer cells and antigen-presenting cells (APCs). This interaction is essential for maintaining immune homeostasis and preventing excessive immune responses. The main functions of CD279 are as follows:

 

1. Immune checkpoint regulation: CD279 is an important regulator of the immune response. When bound to its ligand, CD279 transmits inhibitory signals that suppress T cell activation and proliferation, preventing excessive immune responses. This mechanism is essential to avoid autoimmunity and immune-mediated tissue damage.

 

2. T-cell exhaustion: CD279 is closely linked to T-cell exhaustion, a state of dysfunction commonly observed in chronic infections and cancers. Persistent CD279 signaling can impair the effector function of T cell, leading to ineffective immune responses against persistent pathogens or tumors.

 

3. Immune evasion in cancer: The PD-1 pathway is frequently exploited by cancer cells to evade immune surveillance. Through the upregulation of PD-L1 expression, cancer cells can effectively suppress the cytotoxicity of tumor-specific T cells, thereby permitting tumor growth and immune escape.

 

4. Therapeutic targets: CD279 has emerged as an important target for cancer immunotherapy. Monoclonal antibodies targeting CD279 or its ligands have been developed to block inhibitory signaling and enhance anti-tumor immune responses. Remarkable has been achieved in the treatment of various cancers using drugs such as pembrolizumab and nivolumab.

 

5. Autoimmune Diseases: Dysregulation of CD279 signaling can lead to autoimmune diseases, as excessive suppression of T-cell responses may disrupt immune tolerance. Therefore, a comprehensive understanding and regulating CD279 signaling are critical for the development of therapies for autoimmune diseases.
