CD258 (Gene ID:8740), also known as LIGHT or TNFSF14, is a member of the tumor necrosis factor (TNF) ligand family. It plays an important role in the immune system. Acting as a ligand for TNFRSF14 (also known as HVEM), CD258 exerts influence on various immune responses. It is expressed in activated T cells, NK cells, monocytes, granulocytes, and dendritic cells, thereby contributing to immune response and may influence inflammation and disease processes. The interactions between TNFSF14 and two major receptors-herpesvirus entry mediator (HVEM, also known as TNFRSF14) and lymphotoxin beta receptor (LTβR). These interactions have distinct effects on immunomodulation and lymphoid tissue development:
1. TNFSF14 activates HVEM and LTβR signaling pathways on immune cells. HVEM signaling enhances T-cell activation, cytokine production, and T-cell proliferation, while LTβR signaling is involved in the formation of secondary lymphoid structures such as lymph nodes and Peyer's patches. Thus, TNFSF14 contributes to the activation of immune cells and the development of immune response mechanisms.
2. TNFSF14 can promote the production of pro-inflammatory cytokines and chemokines. This attracts immune cells to sites of infection or tissue damage for pathogen clearance or tissue repair. TNFSF14 plays a crucial role in the development of lymphoid tissues by interacting with LTβR, which helps organize and maintain their structure necessary for immune cell maturation and function.
3. Autoimmune and Inflammatory Diseases: Dysregulated TNFSF14 signaling has been implicated in the pathogenesis of autoimmune diseases and chronic inflammation, including rheumatoid arthritis and inflammatory bowel disease.
