CD47 (Gene ID:961) was initially discovered in the early 1990s as an antigen expressed on human erythrocytes. It is a member of the immunoglobulin superfamily and is known for its interaction with signal regulatory protein alpha (SIRPα) and platelet response protein-1 (TSP-1). Subsequently, CD47 has been acknowledged as a key player in the "don't eat me" signaling pathway, which inhibits macrophages from engulfing healthy cells. The main functions of CD47 are as follows:
1. CD47 acts as a "don't eat me" signal by binding to SIRPα on macrophages, thereby initiating an inhibitory signal to macrophages, preventing them from engulfing and destroying CD47-expressing cells. This mechanism is essential to avoid immune clearance of healthy cells.
2. CD47 plays a role in maintaining tissue homeostasis by inhibiting the phagocytosis of senescent or damaged cells, thereby preserving the integrity of various tissues and organs .
3. CD47 helps regulate angiogenesis, the process of new blood vessel formation. Depending on the environment, its interaction with TSP-1 can either promote or inhibit angiogenesis.
4. Many cancer cells upregulate CD47 as a strategy to evade the immune system. Elevated expression of CD47 on cancer cells can inhibit macrophage-mediated phagocytosis, allowing tumors to evade immune surveillance.
