Peptide PEGylation

We are happy to help you improve the success of your biosimilar discovery research. PEG modification, also known as polyethylene glycol modification, is a method of chemically coupling PEG to protein or peptide molecules to enhance the activity of peptides. Since Davies modified bovine serum albumin with PEG in 1977, PEG modification techniques have been widely used in the chemical modification of many proteins and peptides. PEG modification can prolong half-life, decrease or disappear immunogenicity, reduce toxicity and enhance physical, chemical and biological stability.


Application of Peptide PEGylation:


The application of peptides as biosimilars-lead-compounds in vivo is limited to a certain extent, mainly reflected in the filtration of the glomerulus, hydrolysis and destruction of protease in vivo and antigenic reactions caused by peptides in vivo. The PEGylation of peptides greatly reduced these three constraints and improved their application in living organisms.

Because PEG is hydrophilic, can improve the overall hydrophilic and hydrophobic properties of polypeptide, small molecule PEG can be used as a Link, the modification group and the polypeptide together, sometimes more advantageous than 6-amino caproic acid. A common one is Biotin-PEG4-Peptide.

The PEG modification sites of polypeptides are N-terminal, C-terminal, Lys side chain and the sulfhydryl group of Cys. The molecular weight range of the modified PEG monomer is between PEG2 and PEG24. The molecular weight of the PEG macromolecule ranges from PEG500 to PEG40K.
