Library Screening Services

A compound library or chemical library is a collection of stored chemicals that are usually ultimately used for high-throughput screening or industrial manufacturing. Depending on their scope (specific or diverse targets) and design (chemical space or scaffold), compound libraries can be classified as key bioactive libraries, natural product libraries, fragment libraries and diversity sets. In the early stages of projects in the last decade, it was common practice to screen large numbers of compounds in high-throughput analyses that uniformly covered a large fraction of the chemical space (a diversity-driven paradigm) to identify those with the potential to modulate the target of interest. The high cost of such large-scale screens has necessitated the use of regression analysis optimized primarily for size and speed, and the increasing realization that the drug characterization space is far from random has recently led to the use of smaller, higher-quality screening pools (target-driven approaches). All of these compound libraries and their screening methods have their specific advantages and disadvantages.

 

Inhibitor Screening Steps:

 

Screening is performed in three steps, including a first screen, a second screen, and IC 50 determination. First, the inhibition rate of the compounds is determined.  Each compound is placed in 4 wells of a 384-well plate and the other wells are used for positive and negative controls. The compounds are placed randomly on the plate so that a group of extracted compounds will not be placed alongside each other on the plate. The average of four inhibition rates for each compound was compared to the first screened criteria. These criteria include an inhibition rate of >25% and an inhibition rate greater than the average value plus three times the standard deviation of the plate of the assayed compound, where the observed inhibition rates of the positive and negative controls are not taken into account. Next, the inhibition rate of the screened compounds is measured on a plate using the same procedure as for the first screen, where the compounds are dissolved from the fresh powder. The IC 50 values of the screened compounds are then assessed.

Based on the ForteBio Octet platform, KMD Bioscience performs high-throughput screening of screened compounds with target proteins, along with cellular validation (cell proliferation, cytotoxicity, target inhibition rate, etc.), avoiding false positives, and using MS and NMR screening to determine the structure of the compounds, allowing for their optimization.
