Protein isolation and purification is an essential part of proteomics research. KMD Bioscience has many years of experience in the purification and isolation of native proteins from tissue fluids, saliva, intestinal fluids, animal or human serums, and even some animal tissues, etc. Our scientists will analyze the approximate molecular weights of the natural proteins provided by our customers, buffer compositions, and background information to make specific analyses and feasible solutions.
KMD Bioscience has established a variety of protein purification systems (affinity chromatography, ion exchange, molecular sieve, hydrophobic chromatography, HPLC, nickel affinity chromatography, etc.) to choose from, providing strong support for customers' downstream experimental research. Our advanced AKTA purification system and a variety of specifications of pre-packed purification columns can be used to purify different sources of natural proteins. What’s more, more than a dozen types of GE purification matrixes have been developed to enable simultaneous large-scale protein purification, saving customers' research time and meeting customers' needs.
Native Protein Purification Service
The isolation and purification of native proteins are generally divided into three types: isolation and purification of natural large molecule proteins, isolation and purification of natural small molecule proteins, and isolation and purification of mixed unknown protein samples. Since natural proteins do not contain any affinity tags, it is difficult to meet the demand for simple one-step purification. We will mainly use ion exchange and molecular sieve as the core of the purification of natural proteins.
Fig 1. Native proteins purification
Separation and purification of natural large molecule proteins: According to the physicochemical properties of the target proteins to be purified, we will use molecular sieves with appropriate pore size for separation and purification, and then according to the pH value of the sample buffer we will use appropriate anion and cation adsorption columns to identify and collect the corresponding elution peaks.
Separation and purification of natural small molecule proteins: Firstly, remove the large molecule impurities through molecular sieve multilayer separation and filtration, then purify the small molecule compounds by using macroporous resin or inverse resin, and further identify the small molecule proteins by HPLC and MS. Due to the low content of small molecule proteins, the amount of purified proteins will be relatively small.
Separation and purification of mixed unknown protein samples: KMD Bioscience will formulate a corresponding plan for separation and purification according to the characteristics of the target proteins to be separated, and finally confirm the separated products through mass spectrometry, activity identification and other methods.
