Rabbit Single B Cell Screening Service

KMD Bioscience has been working on antibody development for over 9 years. Equipped with an experienced antibody production team, improved technical procedure, and advanced antibody production equipment, KMD Bioscience can provide customers with high-quality recombinant monoclonal antibody production services, including rabbit source, mouse source, camel source, bovine source, etc. Based on the single B cell platform, KMD Bioscience can provide rabbit monoclonal antibodies that meet the requirements of downstream experiments with good specificity and high affinity.

Rabbit Monoclonal Antibody Advantage:

High Specificity and Affinity: Rabbit monoclonal antibodies tend to exhibit higher specificity and affinity for their target antigens compared to mouse monoclonal antibodies. This is attributed to the rabbit immune system's ability to generate a diverse repertoire of antibodies with enhanced recognition capabilities.

Broader Epitope Recognition: Rabbits have a more diverse immune system than mice, allowing them to recognize a wider range of epitopes on target antigens. Rabbit monoclonal antibodies can therefore bind to epitopes that may be inaccessible to mouse monoclonal antibodies, increasing the likelihood of successful antigen detection.

Reduced Background Noise: Rabbit monoclonal antibodies often have lower background noise in immunohistochemistry (IHC), immunofluorescence (IF), and other applications compared to mouse monoclonal antibodies. This results in clearer and more reliable staining patterns, particularly in tissues with high endogenous mouse IgG levels.

Improved Signal Intensity: Due to their higher affinity and specificity, rabbit monoclonal antibodies typically generate stronger and more consistent signals in immunoassays such as Western blotting and ELISA. This enhanced signal intensity allows for improved detection sensitivity and reproducibility.

Increased Stability: Rabbit monoclonal antibodies tend to be more stable than mouse monoclonal antibodies, exhibiting longer shelf life and retaining their activity under a wider range of storage conditions. This increased stability reduces the need for frequent antibody optimization and minimizes experimental variability.

Compatibility with Multiple Species: Rabbit monoclonal antibodies are highly cross-reactive and can be used in a wide range of species, including humans, mice, rats, and others. This versatility makes them suitable for diverse research applications and facilitates comparative studies across different model organisms.

Less Immunogenicity in Humans: Rabbit monoclonal antibodies are less likely to elicit an immune response in humans compared to mouse monoclonal antibodies. This reduces the risk of interference with experimental results and minimizes potential complications in therapeutic applications.

Rabbit Single B Cell Screening Background:

Monoclonal antibodies are among the most significant biological tools used in medicine and biology that have revolutionized the field of diagnostics, therapeutics, and targeted drug delivery systems for many diseases. Existing monoclonal antibody development technologies, such as hybridoma technology, suffer from a loss of diversity due to the efficiency of cell fusion, and phage display technology produces antibodies whose heavy and light chains are not naturally paired. For these reasons, traditional techniques are inefficient, and considerable time is required to develop antibodies.

During the past few decades, different techniques have been developed to produce monoclonal antibodies. Single B cell cloning technology offers many advantages compared to other methods and has been used to generate monoclonal antibodies from different species including rabbits. Compared with mouse monoclonal antibodies, rabbit monoclonal antibody is widely used in scientific research because of their high affinity and abundant epitopes. Based on the single B cell cloning technique, positive B cells can be obtained from peripheral blood directly after animal immunization and antibody gene sequences can be cloned. Thousands of target-binding positive B cell clones can be obtained from a single immunized animal, and the resulting rabbit monoclonal antibody has the outstanding advantages of high specificity, high activity, and high affinity.

KMD Bioscience has been working to refine its technology and has built up an independent rabbit single B cell sorting platform. Taking advantage of the rabbit immune system, KMD Bioscience obtains rabbit B lymphocytes with high specificity and high affinity. Screening B lymphocytes to obtain antibody genes and sequences and obtains rabbit monoclonal antibodies through recombinant expression, which has a high success rate and superior quality.

Rabbit Single B Cell Screening Service Process:

Rabbit Single B Cell Screening Service Workflow:

Step1: Rabbit Immunization

* A Rabbit was immunized 4-7 times

* Serum titers were measured by ELISA and collected peripheral blood

9-10 weeks

Step2: B cell enrichment and screening

* Collection of lymphocytes or PBMCs by density gradient centrifugation from a few milliliters of animal blood

* Fluorescent staining of plasma cells with endoplasmic reticulum (ER)-tracker

* Sorting of stained cells by fluorescence-activated cell sorting (FACS)

* Adsorption of cells binding to the antigen using antigen-coated magnetic beads

* Separation of one cell per 10 µL into a 384-well plate

* Selection and confirmation of cell-bead complexes under an inverted phase-contrast microscope

Step3: Single B Cell Sequencing

* Reverse transcription using gene-specific primers and SuperScript IV reverse transcriptase

* First PCR, using primers annealing to the signal peptide (SP) sequence and constant region of antibody genes

* Second PCR, using primers with tails required for the subsequent Gibson assembly step

* Gibson assembly to combine T7 promoter and terminator with antibody genes

* PCR to amplify DNA fragments for cell-free protein synthesis. HA and His tags are present downstream of leucine zipper A (LZA) and B (LZB), respectively

* High-throughput sequencing

Step4: Antibody table and purification

* Antibody small transfection or amplification
