ELISA (Enzyme-linked Immunosorbent Assay), based on immunological reaction, combines the specific reaction of antigen and antibody with the efficient catalytic effect of the enzyme on the substrate. It is based on the adsorption of a known antigen or antibody onto a solid-phase vector, so that the enzyme-labeled antigen-antibody reaction can be carried out on the surface of the solid-phase, and the antigen or antibody can be measured qualitatively or quantitatively. The solid-phase vector is a polystyrene microtitre plate, and the reaction of antigen and antibody is carried out in the wells of the titer plate, and after incubation with each reagent, the excess free reactants can be removed by washing, to ensure the specificity and stability of the test results. Therefore, ELISA assay has the advantages of high sensitivity, rapid detection, high specificity and good reproducibility, and is widely used in many fields such as immunodiagnosis, detection and biology.  Experimental Principle

* The principle of ELISA is based on the solid-phase solidification of antigens or antibodies and the enzymatic labeling of antigens or antibodies. In the assay, the test specimen (in which the antibody or antigen is measured) reacts with the antigen or antibody on the surface of a solid-phase vector. The antigen-antibody complex formed on the solid phase vector is separated from other substances in the liquid by washing. Enzyme-labeled antigen or antibody is then added, which also binds to the solid-phase vector by the reaction. At this point, the amount of enzyme in the solid phase is proportional to the amount of the substance being examined in the specimen. After adding the substrate to the enzyme reaction, the substrate is catalyzed by the enzyme to become a colored product, and the amount of the product is directly related to the amount of the substance examined in the specimen, so it can be analyzed qualitatively or quantitatively according to the shade of the color.

Application

* Detects the concentration of target proteins in samples by using antigen-antibody reaction, commonly used in the detection of secreted proteins.

* The concentration of target proteins in the sample can be quantitatively calculated by using the same batch of test standards to obtain the content of target proteins in each experimental group.

* Can be used to determine intermolecular interactions, such as protein-protein and protein-nucleic acid interactions.

Sandwich ELISA assays are commonly used to detect large molecules of antigen or antibody and are called double-antibody sandwich assays because the target is sandwiched between a capture antibody encapsulated on the well and a detection antibody. They are generally categorized as double antibody sandwich assay, which is used to detect the antigen, and double antigen sandwich assay, which is used to detect the antibody.

 Experimental Principle

*Take the double antibody sandwich method as an example: an antibody with specificity is coated on an enzyme plate and added to the sample to be tested. If the sample contains the antigen to be tested, it will bind specifically to the antibody coated on the enzyme plate, and then another antibody directed against the antigen to be tested is added, which is usually called the detection antibody. The detection antibody is usually labeled with HRP enzyme, and then TMB, the substrate of HRP enzyme, is added. TMB is catalyzed by the enzyme and undergoes a color reaction; the more antigen in the sample, the darker the color will appear.

*This method is mainly applicable to the qualitative or quantitative detection of antibodies, antigens and proteins.
