CD275 (Gene ID:23308), also known as ICOSLG (inducible T-cell co-stimulatory molecule ligand), is a gene located on human chromosome 21. It is a member of the B7 family of co-stimulatory molecules and is essential for immune system function. CD275 is a glycosylated transmembrane structural protein. The interaction between ICOSLG and its receptor, ICOS, is essential for T-cell activation, proliferation, differentiation, and cytokine production, as well as antibody secretion from B-cells during the secondary immune response. This response plays a key role in immune responses, including immunopathogenesis associated with infection, hypersensitivity reactions, autoimmune diseases, transplantation immunity, and tumor immunity.
ICOSLG acts as a prominent co-stimulatory factor in endothelium-mediated T cell activation and plays an important role in the reactivation of effector/memory T cells on the endothelium, regulating the entry of immune cells into inflamed tissues. The gene is expressed in both non-lymphoid and lymphoid tissues. While its expression is usually low in initial B cells and monocytes, it can be rapidly upregulated in response to IFN-γ, TNF-α, or LPS stimulation. This upregulation mainly regulates the secretion of Th2 cytokines, which influences the type of immune response.
