TNFSF18 (tumor necrosis factor superfamily member 18) (Gene ID: 8995), also known as GITRL (glucocorticoid-induced tumor necrosis factor receptor ligand), is a gene encoding a cell-surface and soluble protein belonging to the tumor necrosis factor (TNF) superfamily. TNFSF18 plays a crucial role in the regulation of immune responses and is involved in various aspects of immune functions. Originally identified as a molecule induced by glucocorticoid therapy, TNFSF18 is expressed by a variety of immune cells including dendritic cells, B cells, and macrophages, exerting its influence on immune regulation, particularly within the context of T cell responses. The major functions of TNFSF18 are as follows.
1. T-cell activation: The primary function of TNFSF18/GITRL is to regulate T-cell activation. It interacts with its receptor GITR (Glucocorticoid Induced Tumor Necrosis Factor Receptor), which is expressed in T cells. GITRL-GITR interaction enhances T cell proliferation and cytokine production, contributing to an effective immune response.
2. Immune tolerance: TNFSF18 also has immunomodulatory functions. In some cases, it induces regulatory expansion of T cell (Treg) and suppresses the immune response. This dual mechanism of enhancing and suppressing T cell responses is contingent upon the microenvironment and specific immune signals.
3. Anti-tumor immunity: The potential of GITRL as a target for cancer immunotherapy has been investigated. Modulation of GITRL-GITR interactions can enhance anti-tumor immune responses by enhancing T cell activity against tumor cells.
Autoimmune diseases: Dysregulation of the GITRL-GITR signaling is associated with autoimmune diseases. Inhibition of this pathway has been explored as a potential therapeutic approach to control autoimmune responses.
