The CD274 gene (Gene ID:29126), also known as programmed death ligand 1 (PD-L1) or B7 homologue 1 (B7-H1), is critical in immunoregulatory responses. It encodes a type I transmembrane protein weighing 40 kDa, PD-L1, which plays an important role in suppressing the adaptive immune system during pregnancy, tissue allografts, autoimmune disorders, and hepatitis, among other conditions. PD-L1 was initially characterized as an immunomodulatory molecule in 1999 at the Mayo Clinic. Due to its ability to aid tumor cells in evading antitumor immunity, it has been identified as a cancer-associated factor. PD-L1 interacts with its receptor, PD-1, and is expressed on activated T cells, B cells, and myeloid cells to regulate their activation or suppression. This interaction inhibits T-cell receptor (TCR)-mediated activation of IL-2 production and T-cell proliferation and plays a key role in immune response regulation.
The expression of PD-L1 is influenced by interferons. For example, IFN-γ induces the expression of PD-L1 in a variety of cell types, including T cells, NK cells, macrophages, and epithelial cells. The PD-L1 gene promoter contains a response element for the interferon regulatory factor IRF-1, highlighting the regulation of the gene by immune signaling.
Interestingly, the expression of PD-L1 varies across cell types and under different conditions. For example, it is significantly expressed on macrophages and its expression is influenced by factors such as LPS and interferon-γ. This expression is regulated by components such as STAT1 and can exhibit variability based on the activation status of the macrophage.
