TIGIT (Gene ID:201633) is a member of the poliovirus receptor (PVR) family, also known as WUCAM, VSTM3, VSIG, and is a receptor of the Ig superfamily. TIGIT is expressed on natural killer (NK) cells and T cells, featuring an immunoglobulin-like structural domain, transmembrane region, and an immunoreceptor tyrosine-based inhibitory motif (ITIM). TIGIT alters the secretion of IL-12 and IL-10 by competitively binding to CD155 on dendritic cells (DCs), and also directly inhibits T cell and natural killer cell function. TIGIT is emerging as an important immune checkpoint due to its ability to inhibit the killing of tumor cells by NK cells and T cells, which plays a key role in limiting both adaptive and intrinsic immunity to tumors.
