HER1 (Gene ID:1956), also known as EGFR (epidermal growth factor receptor), is a gene that encodes a cell surface protein belonging to the ErbB family of receptor tyrosine kinases. EGFR plays a critical role in cell growth, differentiation, and tissue development. Initially identified as a cell surface receptor binding epidermal growth factor (EGF) and related ligands, EGFR is widely expressed in a variety of tissues, including skin, lung, and gastrointestinal tract. Dysregulation of EGFR signaling has been linked to cancer, leading to extensive research on its role in cancer biology. Activation of EGFR initiates intracellular signaling pathways that promote cell growth and proliferation by regulating processes such as cell cycle progression and DNA synthesis essential for normal tissue development and repair. EGFR signaling is associated with tissue differentiation and development, especially in skin and epithelial tissues. It serves to maintain tissue integrity and repair. Dysregulation of EGFR is a hallmark of several cancers, including lung, colorectal, and head and neck cancers. Mutation or overexpression of the EGFR gene leads to uncontrolled cell growth, tumor formation, and metastasis. EGFR has emerged as a target for cancer therapy. Drugs known as EGFR inhibitors, such as gefitinib and cetuximab, impede the activation of EGFR and are used to treat certain cancer types.
