CD80 (Gene ID:941), also known as B7-1 or CD28 ligand, is a cell surface protein encoded by the CD80 gene, which is a member of the B7 family of immune checkpoint molecules that play a key role in the regulation of immune responses. In the early 1990s, CD80 was identified as a molecule that is expressed on the surfaces of antigen-presenting cells (APCs), such as dendritic cells, macrophages, and B cells. It interacts with the receptors CD28 and CTLA-4 (CD152) on T cells to regulate T cell activation and differentiation. CD80 is a key co-stimulatory molecule that interacts with CD28 on T cells. This interaction provides a second signal during T-cell activation, as well as T-cell receptor (TCR) signaling. Binding of CD80 to CD28 enhances T-cell proliferation, cytokine production, and effector function, thereby ensuring an effective immune response. CD80 also plays a role in immune tolerance. When T cells encounter APCs expressing CD80 without sufficient activation of the TCR, it can result in either unresponsiveness (unresponsive state) or induction of regulatory T cell (Tregs). These mechanisms effectively prevent undesired immune responses to self-antigens. Moreover, CD80 exhibits a dual role in immunoregulation. Although it provides co-stimulation through CD28, it can also interact with the inhibitory receptor CTLA-4 (CD152) on activated T cells. By competing with CD28 for binding to CD80, CTLA-4 inhibits T cell activation and contributing to immune homeostasis.
