The CD3E gene (Gene ID:916) is a component of the CD3 complex, which consists of CD3E, CD3D and CD3Z (also known as CD3-γ, δ and ζ, respectively). These proteins are associated with the TCR and are essential for the function of T cell . In particular, CD3E acts as the ε-chain within the CD3 complex. CD3E is involved in the development of T cells by being expressed early in T cell precursors and facilitating assembly and surface expression of the TCR complex. Without functional CD3E, normal T cell development is impaired. Furthermore, it is essential for antigen recognition by forming a complex with CD3D and CD3Z to complete the overall structure of the TCR complex. This complex binds to antigenic peptides presented by major histocompatibility complex (MHC) molecules on antigen-presenting cells (APCs). CD3E serves as a target for immunotherapy, particularly in the development of bispecific antibodies (BsAbs) and chimeric antigen receptor (CAR) T-cell therapies. BsAbs conjugated with CD3E and tumor-specific antigens redirect T cells to attack cancer cells, while CAR T cells modified with CD3E-specific receptors exhibit promise for a wide range of applications in cancer therapy.
