HAVCR2 (Gene ID:84868), also known as TIM-3 (T-cell immunoglobulin and mucin structural domain-containing protein 3), encodes a cell-surface protein that is expressed predominantly on immune cells. TIM-3 plays a pivotal role in regulating the immune response and has been implicated in a variety of immune-related disorders. Initially discovered as a marker on T-cells, TIM-3 has subsequently been detected on other immune cell types, including dendritic cells and natural killer cells. TIM-3 has gained significant attention due to its role in immune checkpoint signaling in the context of T cell exhaustion and immune tolerance. TIM-3 is an immune checkpoint receptor that plays a key role in the regulation of T cell responses by interacting with its ligands, such as galactose lectin-9 and phosphatidylserine (PtdSer), to transmit inhibitory signals. The upregulation of TIM-3 on T cells is involved in processes that lead to T-cell exhaustion, inhibiting their effector functions. This phenomenon has been implicated in chronic infections (e.g., HIV and hepatitis), cancer, and autoimmune diseases. As a result, TIM-3 has emerged as a target for cancer immunotherapy. Antibodies that block TIM-3 interactions can enhance anti-tumor immune responses by preventing T-cell exhaustion and promoting cytotoxic T-cell activity. TIM-3 has been implicated in autoimmune diseases such as multiple sclerosis and rheumatoid arthritis. Dysregulation of TIM-3 signaling leads to uncontrolled immune activation.
