CD40 (Gene ID:958) was initially discovered in the late 1980s and serves as an important component of the immune system. It was named after cluster 40 at the First International Workshop on Human Leukocyte Differentiation Antigens. CD40 interacts with its ligand, known as the CD40 ligand (CD40L or CD154), which is expressed primarily on activated T cells. This interaction is critical for the activation of B cells and their subsequent involvement in the immune response. CD40 plays a pivotal role in the activation of B cells, and when CD40 on a B cell binds to CD40L on an activated T cell, it triggers a series of intracellular signaling events in the B cell. This leads to the activation, proliferation, and differentiation of B cells into plasma cells that secrete antibodies. The signaling mediated by CD40 is critical for the formation and maintenance of the germinal centers of secondary lymphoid organs. Germinal centers are specialized microenvironments where B cells undergo affinity maturation and extensive somatic hypermutation, leading to the production of high-affinity antibodies.
