CD134 (Gene ID:7293) is a member of the tumor necrosis factor (TNF) superfamily and represents an important class of T-cell co-stimulatory molecules, also known as OX40 and TNFRSF4, predominantly expressed on T-lymphocytes. CD134 encodes a protein containing 277 amino acids with a molecular weight of 29.34 kDa, and contains three complete cysteine-rich structural domains and one slightly shorter cysteine-rich structural domain in its extracellular region. The extracellular region contains three complete cysteine-rich domains and one slightly shorter cysteine-rich domain. CD134, in combination with its ligand CD252, transduces co-stimulatory signals and participates in a variety of physiological responses between T cells and lymphocytes as well as non-lymphocytes, which are essential for cell survival. Moreover, the binding of CD137 and its ligand CD252 also facilitates augmentation of immune system responsiveness and enhancement of anti-tumor activity. Thus, agonists of CD134 can enhance the efficacy of other cancer therapies such as chemotherapy and checkpoint inhibitors while potentially finding applications in autoimmune diseases and allergies.
