CD273 (Gene ID:80380), also known as PDCD1LG2 or PD-L2, is a protein encoded by the PDCD1LG2 gene and belongs to the family of immune checkpoint receptor ligands. It plays an important role in the negative regulation of adaptive immune responses and is expressed in specific subset of dendritic cells, hepatocytes, and certain macrophages. Additionally, it is involved in the stimulation of dendritic cells and other immune responses. PD-L2 was initially identified in macrophages and dendritic cells, and its overexpression has been significantly observed in a variety of human cancers, including head and neck squamous cell carcinoma (HNSCC), lung squamous carcinoma (LUSC), renal cell carcinoma (RCC), pancreatic ductal adenocarcinoma (PDAC), and cervical cancer. Its high expression in tumor cells is associated with a poor prognosis in certain cancers such as HNSCC, adenoid cystic carcinoma and esophageal cancer. Functionally, PD-L2 can significantly promote tumor progression and immune evasion. It exhibits a higher affinity for PD-1, a key immune checkpoint receptor when compared to another ligand of PD-1. This strong interaction inhibits cytokine secretion and T-cell proliferation, underscoring the importance of PD-L2 in the mechanism of immune escape, PD-L2 is also implicated in processes beyond immune function, such as facilitating cancer invasion and contributing to chemoresistance. In terms of regulatory mechanisms, PD-L2 expression is affected by various factors at the genomic, transcriptional, post-transcriptional, translational and post-translational levels. Genomic amplifications and mutations can lead to increased PD-L2 expression in several cancers. For instance, amplification of the chromosome 9p24.1 region where the PD-L2 gene is located has been observed across various tumor types, enhancing the transcriptional and immunological activity of PD-L2.
