CD172a belongs to the immunoglobulin superfamily of membrane proteins, also known as SIRPα (Gene ID:140885), PTPNS1, MFR, p84, BIT, and SHPS-1. It exhibits predominant expression in myeloid cells, such as macrophages. The protein encoded by the human CD172a gene contains 504 amino acids with a molecular weight of 54.97 kDa. The structure of the protein is complex, with the intracellular region containing four immunoreceptor tyrosine inhibitory motifs and the extracellular region containing three Ig-like regions. The extracellular Ig-like regions contain two IgSF structural domains and an amino-terminal structural domain, which is capable of binding to CD47, a signaling molecule that prevents phagocytosis by macrophages. Interaction between CD172a on the surface of macrophages and CD47 signals macrophages to inhibit their phagocytosis. Therefore, blocking the binding of CD172a to CD47 can promote phagocytosis of tumor cells by macrophages, thus achieving the purpose of anti-tumor.
