CD270, also known as TNFRSF14 (Gene ID:8764) and ATAR (another TRAF-related receptor), is a member of the TNF receptor superfamily. This protein plays a key role in immune regulation, especially in T cell activation. It interacts with a variety of ligands such as HVEM and plays a negative regulatory role on a broad range of immune responses. CD270 plays a central regulator of apoptosis, especially in the immune system. Binding of TRAIL or Apo2L to CD270 induces formation of the death-inducing signaling complex (DISC), leading to caspase activation and ultimately cell death. This process is essential for eliminating infected or damaged cells, preventing the spread of pathogens and the development of cancer.
In addition to its role in apoptosis, CD270 has been implicated in immune regulation, exerting regulatory effects on the activity, survival, and function of immune cells such as T cells and natural killer (NK) cells. The signaling mediated by CD270 promotes cell survival or triggers apoptosis, a process that depends specifically on the cellular environment and the presence of other signaling molecules.
CD270 has received much attention as a potential target for cancer therapy. Some cancer cells overexpress CD270, making them susceptible to apoptosis induction by TRAIL-based therapies. Various agonistic antibodies and recombinant TRAIL proteins targeting CD270 have been developed to selectively induce cancer cell death. Clinical trials are currently underway to evaluate the efficacy of these therapies for various types of cancer.
