Mesothelin, encoded by the MSLN (Gene ID:10232) gene, is a cell-surface glycoprotein expressed primarily in mesothelial cells of the pleura, pericardium, and peritoneal lining. Due to its association with specific cancers, particularly mesothelioma, it is considered a target for cancer therapy. Mesothelin was identified in the early 1990s as a highly expressed cell surface antigen in mesothelial tissue. Its nomenclature reflects its tissue origin. Subsequent investigations have elucidated its role in cancer biology and its role as a tumor-associated antigen. Mesothelin is commonly overexpressed in a variety of cancers, including mesothelioma, pancreatic cancer, ovarian cancer, and lung adenocarcinoma. It has been extensively investigated as a tumor marker for diagnostic and prognostic purposes. Elevated levels of soluble mesothelin-related peptide (SMRP) in the blood have been associated with several cancers. Although the precise role of mesothelin in cancer remains incompletely understood, it is generally recognized that it contributes to cancer progression by promoting cell survival, invasion, and metastasis. Mesothelin's role in these processes makes it an important target for cancer therapy. Mesothelin has become a target for cancer immunotherapy and antibody-drug combination therapy. Monoclonal antibodies against mesothelin, such as Amatuximab and LMB-100, have been developed for the treatment of mesothelin-expressing cancers.
