Discovered in the early 1980s, CD28 (Gene ID:940) was among the pioneering co-stimulatory molecules identified in the immune system. It is a member of the immunoglobulin superfamily and is expressed predominantly on the surface of CD4+ and CD8+ T cells. CD28 is a receptor for CD80 (B7-1) and CD86 (B7-2), which are expressed on antigen-presenting cells (APCs), such as dendritic cells, macrophages, and B cells. The interaction between CD28 and these ligands provide a critical second signal for T cell activation.
CD28 is a key co-stimulatory receptor protein expressed primarily on the surface of T cells and plays a critical role in the regulation of adaptive immune responses. The gene is located on chromosome 2q33.2 and is essential for T cell activation and immune cell interactions. The gene functions of CD28 are as follows.
1. T cell activation: The binding of CD28 to CD80 or CD86 on APCs provides a co-stimulatory signal that enhances the activation of T cell. This signaling is indispensable for complete activation of the initial T cells, as activation of the T cell receptor (TCR) alone is usually insufficient to initiate a robust immune response.
2. Cytokine production: The signaling of CD28 plays a key role in the production of a variety of cytokines, including interleukin-2 (IL-2), which is essential for T cell proliferation and effector function. IL-2 promotes the expansion of antigen-specific T cell populations.
3. T cell differentiation: The signaling of CD28 affects T cell differentiation and promotes the development of effector T cells with specific functions, such as cytotoxic T cells, helper T cells and memory T cells.
