CD272, also known as BTLA (Gene ID:151888), is a protein encoded by the BTLA gene located on the third human chromosome and belongs to the CD28 immunoglobulin superfamily (IgSF). BTLA is a co-suppressor receptor that is expressed in B cells, T cells, macrophages, dendritic cells, NKT cells, and NK cells. BTLA plays a key role in regulating the immune response as a negative regulator of lymphocyte activation and/or function. CD272 is a recently discovered immune checkpoint receptor that regulates the immune response. It is structurally similar to other immune checkpoint receptors such as PD-1 and CTLA-4. Immune checkpoints are essential for maintaining immune system homeostasis and preventing excessive immune responses that may lead to autoimmunity or tissue damage. The primary function of CD272 is to regulate immune responses by inhibiting the activation of immune cells. CD272 achieves this regulatory effect through its interaction with herpesvirus entry mediator (HVEM, also known as TNFRSF14), which acts as its ligand. By binding to HVEM, CD272 inhibits T-cell activation and cytokine production, thereby suppressing the immune response. CD272 signaling also prevents B-cell activation and antibody production.
