CD73 (Gene ID:4907), also known as extracellular-5'-nucleotidase, is a cell surface enzyme encoded by the NT5E gene, which plays an important role in purine metabolism, immunomodulation, and tissue protection. CD73 is a glycosylphosphatidylinositol (GPI)-anchored protein that catalyzes the conversion of extracellular adenosine monophosphate (AMP) to adenosine, a nucleoside known for its potent immunosuppressive and tissue-protective properties. CD73 is widely expressed in a variety of tissues, including the endothelium, immune cells, and the tumor microenvironment. CD73 is a pivotal enzyme in the adenosine pathway, responsible for dephosphorylates AMP generated through ATP catabolism into adenosine. Adenosine acts as a signaling molecule with immunosuppressive properties on immune cells, reducing inflammation and preventing excessive immune responses. Adenosine produced by CD73 plays an important role in immune system by inhibiting immune cell activation through its receptors (A1, A2A, A2B, and A3), suppressing the function of T cells, B cells, natural killer (NK) cells, and macrophages. This mechanism aids in maintaining immune tolerance and prevents autoimmune reactions. Furthermore, adenosine derived from CD73 safeguards tissues from ischemia (hypoxia) and inflammation. It promotes vasodilation, reduces tissue damage during inflammation, and improves blood flow to injured areas.
