CD357/GITR (Gene ID:8784) is a member of the tumor necrosis factor receptor (TNFR) superfamily, initially discovered in the late 1990s. It is expressed predominantly a variety of immune cells, including T cells, B cells, dendritic cells, and natural killer (NK) cells. CD357/GITR is notable for its involvement in immune regulation and its potential as an immunotherapeutic target. CD357/GITR acts as a co-stimulatory receptor on T cells. Upon binding to its ligand GITRL (GITR ligand or TNFSF18), CD357/GITR enhances T cell activation and proliferation. GITRL is normally expressed on APCs. This co-stimulation contributes to the expansion of effector T cell populations. CD357/GITR is critical for the function of regulatory T cells (Tregs), which are a specialized subset of T cells responsible for suppressing suppressing immune responses and maintain immune tolerance. The signaling pathway mediated by CD357/GITR disrupts Treg suppression, leading to activation of effector T cells and enhanced immune responses. CD357/ GITR has received attention as a target for cancer immunotherapy. Activation of agonistic antibodies or ligands targeting CD357/GITR on T cells can enhance anti-tumor immune responses. The potential of combination therapies involving CD357/GITR agonists in cancer treatment is currently being investigated in preclinical and clinical studies.
