CD96 (Gene ID:10225), also known as TACTILE (T cell activation with increased late expression), is a cell surface protein encoded by the CD96 gene. It is a member of the immunoglobulin (Ig) superfamily and plays a role in immune cell interactions and regulation. Initially identified as a T-cell activation antigen exhibiting augmented late expression, hence CD96 earned its name TACTILE. It is widely expressed on immune cells, including T cells, natural killer (NK) cells, and specific subsets of B cells. The involvement of CD96 in immune responses has garnered significant attention. The gene function of CD96 can be described as follows: CD96 is an adhesion molecule that interacts with the ligand CD155 (also known as PVR or Necl-5) on target cells. This interaction plays a role in immune cell adhesion and cytotoxicity, especially in NK cells. Furthermore, CD96-CD155 interaction is involved in the formation of immune synapses between immune cells and target cells. Acting as a co-receptor for NK cells, CD96's binding to CD155 provides activation signals for NK cells to enhance their cytotoxicity against target cells, such as tumor cells or virus-infected cells. CD96 is also expressed on T cells, although less understood than its role on NK cells. It may play a role in T cell activation, adhesion and immune synapse formation. CD96 is an adhesion molecule that plays a role in immune cell adhesion, cytotoxicity, and immune synapse formation. CD96 has the potential to be a therapeutic target for cancer immunotherapy, and is therefore an area of research that has received much attention.
