CD244 (Gene ID:51744) is a member of the signaling lymphocyte activation molecule (SLAM) receptor family, which also includes SLAMF1 (CD150), SLAMF2 (CD48) and SLAMF6 (CD352). These receptors are involved in various aspects of immune cell signaling and interactions.
CD244 plays a key role in regulating NK cell activity and T cell function. Its main functions are as follows:
1. NK cell activation: CD244 functions as an activation receptor on NK cells, capable of inducing NK cell activation upon interaction with its ligand CD48 expressed on target cells. This activation triggers the release of cytotoxic particles containing perforin and granzyme, leading to the lysis of target cells, including infected or cancerous cells. Activation of CD244 also enhances the ability of NK cells to produce cytokines, which further contributes to immune responses.
2. Immune Surveillance: CD244 facilitates NK cells recognize and eliminate abnormal or infected cells. As a co-stimulatory receptor, CD244 enhances NK cell cytotoxicity when it binds to CD48 on target cells. This mechanism is critical for immune surveillance and defense against viral infections and tumor development.
3.T cell regulation: CD244 is also expressed on certain subsets of T cell where it exerts a costimulatory effect. In some cases, the signaling mediated by CD244 leads to reduced activation and cytokine production in T cells, thereby contributing to maintain immune tolerance and prevent excessive immune responses.
