CD70 was discovered in the early 1990s as a ligand for CD27, a member of the TNF receptor superfamily. CD27 is predominantly expressed in various immune cells, including T cells, B cells, and natural killer (NK) cells. In contrast, CD70 is expressed on antigen-presenting cells (APCs) such as dendritic cells and activated B cells. The interaction between CD27 and CD70 is essential for an appropriate immune response. CD70 is a costimulatory molecule that interacts with the receptor CD27 on T cells. This interaction provides crucial signals for T cell activation, proliferation and differentiation. It enhances antigen-specific immune responses, especially within T-cell-mediated immunity. CD70-CD27 interactions also play a role in B cell activation. They support B cell proliferation, differentiation into plasma cells and antibody production. This interaction contributes to the development of an effective humoral immune response. CD70 is involved in generating memory T cells, which are essential for long-term immunity. The signaling mediated by CD70 during T cell that exhibit heightened responsiveness upon re-exposed to the same antigen.
